In Search for a Convenient, Scalable Synthesis of 5-(3,4-Dihydroxyphenyl)-γ-valerolactone (DHPV): a Closer Inspection of a Condensation/Elimination/Hydrogenation Route

Abstract

5-(3,4-Dihydroxyphenyl)-γ-valerolactone (DHPV) is a natural product, which is produced in the human gut by a microbial metabolism of (epi)catechins. Despite this postbiotic metabolite is known for its significant pharmacological potential, there are no reports on its convenient synthesis in multigram quantities. With an aim of finding a convenient source of DHPV for preclinical studies, we have undertaken an effort to verify the potential of a literature method based on a condensation/elimination/hydrogenation sequence. We have reproduced the protocol for both reaction steps and identified bottlenecks that may be addressed to further improve the proposed pathway. These findings constitute a solid background for finding a suitable way to obtain DHPV for its further pharmacological evaluation.
