COCRYSTALS - A NEW DRUG FORM

Abstract

Cocrystallization was already known 160 years ago. Cocrystals are structurally homogeneous crystalline materials containing two or more components present in definite stoichiometric amounts. The components are discrete neutral molecular species which are solids at ambient temperature. The use of cocrystallization to generate new forms of active pharmaceutical ingredients (API) was initiated only at the end of the last century. In recent years, the number of publications on this topic has increased significantly, mainly because of the possibility of replacing poorly soluble salts of biologically active substances with their cocrystals. Of particular importance are the physicochemical properties of cocrystals which, when modified, alter the pharmacodynamic parameters of the cocrystallized drug. It is possible to improve the solubility, stability, and – in particular – the bioavailability of drugs.
