MULTITARGET DRUGS – A NEW PARADIGM IN DRUG DESIGN

Abstract

For at least recent 20 years, nearly all medicinal chemistry efforts have been governed by the paradigm "one disease, one gene, one molecular target, one drug". However, our understanding of systems biology and molecular complexity of human diseases substantially shifted current therapeutic thinking towards drugs acting upon many molecular targets, i. e. multitarget drugs. In present paper we discuss theoretical and historical background of intended multitargeting, including benefits of such drugs. The issues of lead-seeking approaches are covered along with the detailed description of pharmacodynamic and pharmacokinetic parameters optimization. We review recent literature in multitarget drugs field and present current conceptual trends.
